16S RNA IN METAGENOMIC ANALYSIS

ABSTRACT

Metagenomics is study of collective genomes of microorganisms. By using metagenomics, genomic analysis of microorganisms will be performed by direct extraction and cloning of DNA from an assemblage of microorganisms, collected from various types of environment such as soil, river, feces and marine. The aim of the study is to examine the 16s RNA in Metagenomic Analysis. In achieving this aim, the following specific objectives were laid out to perform genomic analysis of microorganisms in water, assess rapidly the environmental health condition and status in the future application, evaluate the sequence of information in assessing river water quality and study water quality can widen our perspective in assessing pollution level of the river. In total 24 isolates which are consisted of 18 isolates from polluted rivers and six from unpolluted one, were sequenced. Along these sequences, six of 16S rRNA sequences of colliform bacteria obtained from genebank were placed in Operational Taxonomic Units (OTUs). Phenetic analysis revealed that the river water used in this study were classified into two groups, representing the polluted and unpolluted. Since the tree can clearly distinct unpolluted river water from that of polluted one, we tried to develop a putative sequences motif for both condition. In practical purpose, the sequences motif can be used to screen river water quality in certain river water environment. This study will be of immense benefit to micro biologist, and other researchers who intend to know more on this study and can also be used by non-researchers to build more on their research work. This study contributes to knowledge and could serve as a guide for other study.

CHAPTER ONE

1.1 Introduction

Metagenomics, or also known as environmental genomics, is study of collective genomes of microorganisms. By using metagenomics, we are able to perform genomic analysis of microorganisms by direct extraction and cloning of DNA from an assemblage of microorganisms, collected from various types of environment such as soil, river, feces and marine (Handelsman, 2004; Bhuiyan et al., 2011). The recent development of molecular biology designed to access this wealth of genetic information through environmental nucleic acid extraction has provided a means of avoiding the limitations of culture-dependent genetic exploitation (Cowan et al., 2005).

As a prelude to other parts of this study, this chapter will discuss the background upon which this study was initiated, the statement of problems that led to this study, the Aim and Objectives of the study. Others are Significance of the study, Scope of work, Research hypothesis and questions, Limitations of the Study and Definition of technical terms.

Can the material for “Critical Review of the Role of Community Banking in Rural Development” be used as a guide for Banking and Finance (BF) Project?

Yes it can be used — This research work scrutinized the roles of community banks in rural development is very essential to complete with other parts of the world. Particularly those from industrial countries who are already in the rural development and have easy access to various type of community bank in rural development. To be able to complete effectively with such rural development, Nigeria community banks must be willing to offer attracter terms. The federal government has and is still working hard to encourage the development of community banking in rural areas. A good step towards this is the promulation of roles of community bank in rural development incentives and miscall on loans provision decree 46, which came up with numerous incentive to community bank in rural development. It’s indicated that the type of maintenance and procedures used will help in preserving historical buildings. The research examines the level of application of Facilities Management Procedures in Historical Building using Tafawa Balewa Square as the Case study. The research adopted a survey design and used self- administered questionnaires that were served on 133 users of the case study. Out of this number 88 were received back. The data obtained was analyzed using descriptive and inferential statistical tool. The study found out that, the maintenance of equipment and systems in line with the manufacturer’s specification is a the procedure best carried …

Can the material for “Comparative Study of Antibacterial Activity of Two Selected Medicated Soap and One Local Black Soap on Staphylococcus Aureus From Wound Infection” be used as a guide for Microbiology Project?

Yes it can be used — The study was carried out to compare the Antibacterial Activity of Two Selected Medicated Soap and One Local Black Soap on Staphylococcus Aureus from Wound Infection. In achieving this aim, the following specific objectives were laid out to determine antimicrobial activity of local black soaps on staphylococcus Aureus from Wound Infection and compare the difference between antimicrobial activities of medicated soaps with that of local black soaps on staphylococcus Aureus from Wound Infection. Identification of the bacteria species was by standard microbiological techniques which included colonial examination, gram staining, and biochemical testing. Minimum inhibitory and minimum bactericidal activities of the soaps were determined by disc agar diffusion method. Black soap has the highest antibacterial activity while branded medicated soap 2 …
